چکیده مقاله:

In this study the genetic polymorphism of growth hormone (GH) gene as a candidate gene in livestock was investigated. Blood samples were randomly collected from 34 Alpine and 42 Saanen goats. DNA was extracted from blood samples and a 365 bp region of exon 5 of the GH genewas amplified by polymerase chain reaction (PCR). PCR products were analyzed using single strand conformation polymorphism (SSCP) method on polyacrylamide gel. The results indicated that there are four (G1, G2, G3 and G4) conformational patterns with frequencies 0.38, 0.21, 0.23 and 0.18 for Alpine goat and 0.48, 0.21, 0.17 and 0.14 for Saanen goat, respectively. These results revealed that GH gene was polymorph in this studyand showed that PCR-SSCP is an appropriate tool for detecting polymorphism and evaluating genetic variability.

Cloning, sequencing, and polymorphisms of the Wistar-Imamichi rat growth hormone gene using PCR-SSCP.

We successfully cloned the Wistar-Imamichi (WI) rat growth hormone gene (GenBank accession: GQ890681), which contained 5 exons and 4 introns. Using the polymerase chain reaction-single-strand conformation polymorphism (PCR-SSCP) technique, a novel missense substitution single nucleotide polymorphism was identified and tested for Hardy-Weinberg equilibrium with the χ(2) test. This gene fragment ...

RT-PCR MEDIATED CLONING OF HUMAN GROWTH HORMONE GENE AND I TS EXPRESSION IN E. coli

Human growth hormone (hGH) genomic sequence containing 5 exons and 4 introns was cloned in pcDNA-3 and the constructed plasmid was subsequently used for transfection ofNlli-3T3 cell line using lipofection technique. Expression of hGH in stably transfected cells was assayed using ELISA. Total RNA was extracted from transfected cells and hGH cDNA was amplified by RT-PCR using specific primers...

PCR-SSCP Analysis of Genetic Variation in DNMT Gene Family in Holstein Cattle

Members the DNA Methyltransferases (DNMT) gene family have been shown to play fundamental roles in regulating embryonic growth and development from embryonic fertilization to postnatal life; through regulating the establishment and/or maintenance of specific epigenetic marks. The present study was conducted to identify potential reported mutations within the exon 33 of DNMT-1, intron 4 of DNMT-...
